There is a scout in my troop who is looking to become an "assistant den Chief" for a den I am the current chief for. Is this possible, and if so, what are the rules about it?

Never heard of it. One den, one Den Chief.
There are so many den leaders who would love to have a Den Chief, ask him to volunteer for a different den.

Agreed, plenty of Dens around that need Den Chiefs.
